摘    要:目的探讨γ-氨基丁酸A型(GABA—A)受体阻断剂Gabazine对大鼠黑质网状带GABA能神经元放电的影响。方法采用多管微电极细胞外电生理学记录的方法,观察Gabazine对大鼠黑质网状带GABA能神经元放电的影响。结果正常大鼠黑质网状带微量注射Gabazine可使神经元放电频率由(19.7±2.1)Hz升高到(32.0±5.4)Hz,平均升高(40.0±5.0)%,与生理盐水组相比较差异有显著意义(t=3.25,P〈0.01)。结论内源性GABA—A受体可调节黑质网状带GABA能神经元自发放电活动,这为进一步探究GABA—A受体在帕金森病发病中的作用提供了一定的理论依据。

Abstract:Objective To investigate the effects of Gabazine, a gamma-aminobutyric acid A (GABA-A) receptor antagonist, on the firing activity of GABAergic neurons in substantia nigra pars reticulata of rats. Methods By using extracellular elec- trophysiologieal recording, the effects of Gabazine on the firing activity of GABAergic neurons in substantia nigra pars reticulata was observed. Results Microinjection of Gabazine into the substantia nigra pars retieulata could increase the discharge frequency of the neurons from (19.7±2.1) Hz to (32.0 ± 5.4) Hz, being average of (40.0±5.0)%, which was significantly different as compared with that in the normal saline group (t=3.25,P〈0.01). Conclusion The endogenous GABA-A receptors can modulate the spontaneous firing activity of the substantia nigra pars reticulata neurons, which provides a theoretical evidence for further stud- y of the effect of GABA-A on the development of Parkinson's disease.
