葛根素对慢性缺氧小鼠心肌保护作用的实验观察

摘    要:目的探讨葛根素对慢性缺氧小鼠心肌损伤的保护作用.方法建立小鼠慢性缺氧模型50只,随机分为3组:A葛根素注射液治疗组20只、B单纯缺氧非治疗组20只,正常对照组10只.实验2周后,3组小鼠均处死,分别取小鼠新鲜心肌标本,检测琥珀酸脱氢酶(SDH),三磷酸腺苷酶(ATP),乳酸脱氢酶(LDH),并进行光镜、电镜下心肌病理形态及超微结构变化的观察比较.结果 A组心肌酶和心肌超微结构与对照组相比差异均无显著性(P〉0.05);B组SDH,ATP酶减少,LDH升高;心肌间质水肿,心肌细胞线粒体肿胀,肌丝排列紊乱,与对照组比较差异有显著性(P〈0.01);结论慢性缺氧导致心肌酶及超微结构的改变与病情程度成正比;缺氧导致心肌酶学及超微结构的变化,两者比较,心肌酶的改变比超微结构改变更敏感;葛根素注射液具有保护心肌、减轻缺氧后心肌损伤的作用.

Abstract:Objective To investigate the protective effect of puerarin on myocardiumin of anoxia-mice.Methods Established the model with chronic hypoxia in 40 mice.Then devided into 2 groups:A puerarin infection group 20 mice and B without the infection of puerarin 20 mice,In addition normal contyol 10 mice.After two weeks,all the mice worekilled,the Myocardium samples were taken at once and experlmental challenge. Myocardium succinie dehyrogenase(SDH),adenosine trphosphatase(ATP),lactic dehydrogenase(LDH) were measured.Pathologic changes of myo- cardium were observed under the light and electric microscopes.Results In hypoxia mice,SDH and ATP decrease and LDH increase were observed; in addition,the mesenchyma of the myocardium was edematous and mitochondrions of myocardial were enlarged; the arragement of filaments was irreg- ular(P<0.01).However,there was no significant difference in puerarin and normal contyol groups(P>0.05).Conclusion Anoxia causes myo- cardial harm and is correlative positively with the degree of asphyxia; Hypoxia can cause the changes of the enzymohistochemical and morphological of myocardium,the former is more sensitive than the latter;The Puerarin can protects myocardium and improne the function of the heart.
