发酵支原体抗HIV—1免疫致病机理的研究

摘    要:目的:探讨人类免疫缺陷病毒(human immunodeficiency virus,HIV)感染过程中伴随的白细胞介素-10(IL-10)水平升高是否与合并发酵支原体(mycoplasma fermentans,M.fermentans)感染有关以及IL-10对人外周血单个核细胞(peripheral blood mononuclear cell,PBMC)中HIV-1复制的作用。方法:采用不同浓度的M.fermentans PG18株,用ELISA法检测其对正常人PBMC及HIV-1感染人PBMC产生IL-10的诱导活性;同时,以HIV-1p24抗原为指标,用ELISA半定量法检测人重组IL-10(rHuIL-10)在体外对人PBMC中HIV-1复制的作用。结果:M.fermentans能显著诱导正常人PBMC及HIV-1感染人PBMC产生IL-10,与正常人PBMC相比,M.fermentans对HIV-1感染人PBMC产生IL-10的诱导活性更强;用rHuIL-10处理的人PBMC在感染后第6天对HIV-1的复制产生了抑制作用,抑制作用随rHuIL-10剂量增加而加强,感染前2d较感染后1d加入rHuIL-10产生的抑制作用显著。结论:M.fermentans与HIV感染过程中IL-10水平升高有关;IL-10可抑制人PBMC中HIV-1的复制;M.fermentans可能通过产生IL-10参与并维持(acquired immunodeficiency diseade,AIDS)较长的潜伏期。

Abstract:Objective:Our purpose was to determine whether the increase of IL 10 level is associated with coinfection with M.fermentans during human immunodeficiency virus (HIV) infection and the effects of IL 10 on HIV 1 replication. Methods: The inductive activity of various concentrations of M.fermentans PG18 to normal human peripheral blood mononuclear cells (PBMC) and HIV infected human PBMC were detected by a IL 10 ELISA KIT. The inhibitory effect of rHuIL 10 on HIV 1 replication in human PBMC were studied. Results: M.fermentans could induce IL 10 production in normal human PBMC and HIV infected human PBMC in a dose dependent manner, IL 10 level reach a peak after two days incubation and gradually decrease after that time. Human PBMC pretreated 2 days with human recombinant IL 10 inhibited HIV 1 replication in the 6th day after infected by HIV 1 in a dose-dependent manner. The inhibitory effect was more prominent when IL 10 was added 2 days before infection than IL 10 added 1 day after infection. Conclusion: M.fermentans can induce IL 10 production in normal human PBMC and HIV 1 infected human PBMC. M.fermentans is associated with the increase of IL 10 level in HIV infection. The IL 10 exert a suppressive effect on HIV 1 replication in human PBMC. Therefore M.fermentans may involve in the long latency of AIDS by inducing IL 10 production.
