免疫抑制剂FTY720对Hepa1-6肝癌细胞抑制作用的体外研究

目的探讨免疫抑制剂FTY720对小鼠肝癌细胞Hepa1-6的影响。方法培养小鼠Hepa1-6肝癌细胞，将其分为对照组及0．1、1、10、100μg／ml质量浓度组，处理24h及48h。采用MTT法检测细胞增生，流式细胞术测定细胞周期及细胞凋亡。结果小鼠肝癌细胞增生能力在FTY720作用48h后明显受抑，最大抑制率为62．10％；FTY720对肝癌细胞有促进凋亡作用，在1．0～100．0μg／ml质量浓度范围内，凋亡率随药物浓度增加而增加，分别为：4．07％、8．16％、19．84％；FTY720使G1期显著延长。结论FTY720对肝癌细胞的增生有抑制作用，能使G1期阻滞，促进细胞凋亡。

Inhibitory effect of immunosuppressive agent FTY720 on hepatocellular carcinoma Hepa1-6 in vitro experimental study

Objective　To investigate the inhibitory effect of immunosuppressive agent FTY720 on hepatocellular carcinoma Hepa1-6. Methods　Hepa1-6 cells were cultured, and divided into 4 groups, namely control group and 0.1 μg/ml, 10 μg/ml, 100 μg/ml quality concentration groups. The cells were treated by the drugs for 24 to 48 hours respectively. The inhibitory rate of the cells was measured by MTT assay, and cell cycle and cell apoptotic rate were detected by flow cytometry(FCM). Results　The ability of tumor cell growth were inhibited by FTY720 after 48 h. The maximal inhibition rate was 62.10 %. The apoptosis ratio was increased when FTY720 was 0.1-100 μg/ml, and it was 4.07 %, 8.16 %, 19.84 % respectively. FTY720 significantly prolonged cell G1 phase. Conclusion　FTY720 could inhibit the growth of hepatocellular carcinoma, arrest the cell in G1 phase, and increase apoptosis.
