摘    要:目的观察体外培养传代的毛乳头细胞表达干细胞因子(stem cell factor,SCF)情况;并观察SCF对体外培养的毛乳头细胞生长曲线的影响。方法应用免疫组化及流式细胞技术测量不同代次的毛乳头细胞SCF的表达变化情况,采用MTT比色法观察SCF对毛乳头细胞生长增殖的影响。结果低传代培养的毛乳头细胞SCF的表达较强,随着传代次数的增多,表达逐渐减弱。SCF组与对照组相比,细胞生长曲线的停滞期减少,提前进入对数生长期,并且OD值与对照组相比明显升高,差异有统计学意义(P0.05)。结论 SCF可以促进体外培养的人毛乳头细胞的增殖。

Abstract:Objective To investigate the expressions of SCF in different passages of cultured Dermal Papilla Cells (DPCs) and observe the growth of DPCs in vitro with method of SCF. Methods The expression of SCF in different passages of cultured DPCs was investigated by immunochemistry staining and FCM. The growth curve of DPCs in vitro with addition of SCF was assessed by MTT method. Results The expression of SCF was stronger in early passages compared with that in late passages in cultured DPCs. Compared with the control group, the OD value in the SCF group was increased (P<0.05 ). Conclusion It was suggested that SCF could stimulate the proliferation of DPCs.
