霍姆注射液对内毒素致大鼠急性肺损伤时肺血管内皮细胞ICAM-1的影响

摘    要:目的探讨内毒素致大鼠急性肺损伤(ALI)时,肺血管内皮细胞间黏附分子-1(ICAM-1)表达的变化及霍姆注射液(高渗氯化钠羟乙基淀粉40,HH40)的保护作用。方法健康雄性SD大鼠30只,体重250-300g,随机分为3组(每组10只):空白对照组(C组)给予等容量的NS、急性肺损伤组(L组)和HH40组(H组)均经尾静脉注射大肠杆菌脂多糖(LPS,5 mg/kg),1min后以5ml/kg输注生理盐水或HH40于10 min内输完。注射内毒素4h后放血处死大鼠,检测肺湿/干重比(W/D)、支气管肺泡灌洗液(BALF)中蛋白含量、肺组织肿瘤坏死因子-α(TNF-α)含量和髓过氧化物酶(MPO)活性及光镜下病理变化,采用免疫组织化学方法检测肺血管内皮细胞ICAM-1的表达。结果与C组相比,L组和H组肺W/D、BALF中蛋白含量、MPO活性和TNF-α含量显著增高,ICAM-1明显升高(P〈0.01),肺组织出现明显的病理改变;与L组相比,H组各项指标均显著降低(P〈0.01),病理损伤明显改善。结论ALI大鼠肺血管内皮细胞中ICAM-1呈过度表达,HH40可以抑制肺血管内皮细胞ICAM-1的表达,降低肺W/D、BALF中蛋白含量、MPO活性及TNF-α含量,减轻肺损伤病理学改变,从而达到对ALI的早期保护作用。
