低氧对鼠雪旺氏细胞增殖的影响

摘    要:目的观察低氧对鼠雪旺氏细胞体外增殖的影响,为雪旺氏细胞扩增培养探索新方法。方法原代分离培养雪旺氏细胞并鉴定,将培养至第三代的细胞分为3组,分别为对照组,10%02组和3%O2组。对各组细胞记数;测定培养液中氧含量和乳酸浓度及各组细胞的增殖指数。结果随着环境氧浓度的下降,细胞培养液中的氧含量也逐渐下降(P〈0.05),而乳酸浓度逐渐升高(P〈0.05)。低氧组雪旺氏细胞数目比常氧组明显增加,尤以10%O2组更为明显(P〈0.05),3%O2组在低氧1d和2d时增殖指数低于常氧组,低氧4时,增殖指数增加;而10%O2组增殖指数一直高于常氧组(P〈0.05)。结论轻中度低氧有利于鼠雪旺氏细胞的体外增殖。

Effects of hypoxia on the proliferation of Schwann cells in vitro
Abstract:Objective To investigate the effects of hypoxia on proliferation of Schwann cells and find a new way of expansion of Schwann cells in vitro. Methods Normal rat's sciatic nerves were used for the primary culture Schwann cells. Schwann ceils were dissociated and divided into three groups(control, 10 % O2 and 3 % O2)after identification. The numbers of Schwann cells were counted and the proliferation indexes of Schwann cells were detected by flow cytometry. The concentration of lactic acid and oxygen in the culture fluid were tested. Results The lactic acid decreased as the oxygen concentration descended in of cell culture fluid (P〈0.05). The numbers of Sehwann cells in hypoxia group were more than that in contral group, especially in 10% 02 group (P〈0.05). The proliferation indexes of Schwann cells exposed in hypoxia(3%O2) for one and two days were less than that in contral. It was so until the third day in hypoxia(3%O2).While The proliferation indexes in 10%O2 group were more than that in contral all days(P〈0.05). Conclusion These findings demonstrate that hypoxia, especially mild low oxygen, could promote the proliferation of Schwann cells in vitro.
