甘珀酸钠苦参素包合物对免疫性肝损伤小鼠的影响

摘    要:目的研究甘珀酸钠苦参素包合物(OCSC)对卡介苗联合脂多糖所致免疫性肝损伤小鼠的影响。方法在实验始末给小鼠尾静脉分别注射卡介苗(BCG)和脂多糖(LPS)制备免疫性肝损伤模型,其间用OCSC[100、50、25mg/(kg.d)]连续灌胃给药12d,取血清,用酶联免疫法(ELISA)法检测血清TNF-α、IL-1β、IL-4、IL-10水平,并观察肝脏病理学变化。结果 OCSC100mg/(kg.d)能显著降低免疫性肝损伤小鼠血清中1型辅助T细胞(Th1)TNF-α和IL-1β水平(P〈0.01),升高2型辅助T细胞(Th2)IL-4、IL-10水平(P〈0.01);OCSC可明显减轻肝损伤。结论 OCSC对小鼠免疫性肝损伤具有保护作用,可能是通过调节Th1/TH2细胞的功能状态,使之分泌的细胞因子水平达到平衡而发生作用。

The effect of oxymatrine-carbenoxolone sodium complex on immunogenic hepatic injury in mice

Abstract:Objective To study the influence of oxymatrine-carbenoxolone sodium complex(OCSC)on immunogenic hepatic injury induced by Bacillus Calmette-Guerin and lipopolysaccharide in mice.Methods The mice were treated with Bacillus Calmette-Guerin by vena caudalis injection for establishing immunogenic hepatic injury model.The mice were given different doses of OCSC(100,50,25 mg/kg/d)and biphenyl dimethyl dicarboxylate(200 mg/kg/d)by intragastric administration for 12 days.2 hours after the administration of drugs in final,the mice were injected of lipopolysaccharide by vena caudalis to finish the model making.After 16h,the blood was collected and detected the levels of cytokines such as TNF-α,IL-1β,IL-4 and IL-10 by ELISA,and observed the pathological change in liver.Results Levels of Th1 cytokines(TNF-α and IL-1β)increased in model group but Th2 cytokines(IL-4 and IL-10)decreased(P0.01).Comparing with model group,OCSC(100 mg/kg/d)reduced the levels of TNF-α and IL-1β and increased levels of IL-4 and IL-10(P0.01).OCSC also reduced the hepatic injury significantly.Conclusion OCSC can keep the balance of Th1/TH2 cytokines,reduces the hepatic injury.
