原花青素对氨基脲染毒小鼠肾脏损伤的影响

摘    要:目的:探讨原花青素(PC)对氨基脲(SEM)染毒小鼠肾脏损伤的影响。方法:50只成年昆明种(KM)小鼠,随机分为5组:溶剂对照组,SEM染毒组,SEM染毒+低、中、高剂量PC保护组;溶剂对照组,灌胃去离子水,每天1次,连续6周;SEM染毒组,前两周灌胃56.25 mg/(kg.bw)SEM溶液,后4周灌胃去离子水;低、中、高剂量PC保护组,前2周均灌胃56.25 mg/(kg.bw)SEM溶液,后4周分别灌胃100 mg、200 mg、400 mg/(kg.bw)PC溶液。末次灌胃24 h后,摘眼球采血后处死小鼠,测定相关指标。结果:SEM染毒组小鼠肾脏脏器系数低于溶剂对照组(P<0.05);中、高剂量PC保护组小鼠肾脏脏器系数均高于SEM染毒组(P<0.05);SEM染毒组肾组织形态,可见系膜细胞中、重度增生,肾小管内出现蛋白管型及大量巨噬细胞浸润;中、高剂量PC保护组,肾小管上皮细胞变性及炎性浸润等病理变化明显改善;SEM染毒组小鼠血清UREA、CRE、UA含量高于溶剂对照组(P<0.05),中、高剂量PC保护组UREA、CRE、UA含量均低于SEM染毒组(P<0.05);与溶剂对照组比较,SEM染毒组小鼠肾脏中的SOD活性下降、MDA含量增高(P<0.05),中、高剂量PC保护组SOD活性升高,MD含量降低与SEM染毒组相比,差别均有统计学意义(P<0.05)。结论:原花青素对氨基脲引起的小鼠肾损伤有保护作用,原花青素对氨基脲致小鼠肾损伤的保护机制与抗氧化损伤有关。

Abstract:Objective:To explore the effect of procyanidins on kidney injury induced by semicarbazide in male mice.Methods:50 adult Kunming(KM) mice were equally divided into 5 groups randomly as follows: solvent control group,SEM group,SEM+low-dose PC group,SEM+mid-dose PC group,SEM+high-dose PC group.Solvent control group fed with deionized water and the dose of SEM was fix on 56.25 mg/(kg·bw) but that of PC vary as 100 mg/(kg·bw),200 mg/(kg·bw),400 mg/(kg·bw).Mice of SEM group had been gavaged by 56.25 mg/(kg·bw) for 2 weeks,then continued orally different doses of PC for 4 weeks,at last all animals were further with 56.25 mg/(kg·bw) SEM respectively.The total gavaging volume and freqence were controlled according to 10 ml/(kg·bw) and 1 per day.span of gavaging lasted for 6 weeks.24 h after the last gavages,eyeballs were removed and blood was collected after the mouse were killed.Besides,routine pathological section and HE staining of kidney were operated.Result: The organ coefficient of kidney mice underweight SEM were lower than that in solvent control group(P<0.05);yet that in the medium and high doses of PC protection are higher than SEM group(P<0.05).Kidney pathological Observation revealed that a number of severe disorder associated with ballooning degenerated and inflammatory cell infiltration in SEM group,yet kidney cell degeneration and necrosis reduced significantly in in medium and high doses of PC protection group.Contents of UREA,CRE,UA in serum increased in SEM group(P<0.05),but the activity or content of those in midium and high doses of PC protection ensued decreased(P<0.05),furthermore,and the protective effect of previous protection were more significant(P<0.05).compared with solvent control,Activities of SOD,in kidney of SEM group animals decreased but those of MDA increased(P<0.05).Compared with SEM group,contents in medium and high doses of PC protection ensued increased(P<0.05),furthermore,the protective effect of protection were more significant(P<0.05).Conclusion:Procyanidins(PC) have protection mice the kidney injury induced by semicarbazide(SEM);Relevant of protection mechanism and antioxidant the procyanidins on kidney injury induced by semicarbazide in male mice.
