豚鼠内耳增殖期细胞的研究

摘    要:目的:检测哺乳动物内耳中具有增殖能力的影响。方法:采用增殖细胞核抗原(PCNA)单克隆抗体,以免疫组化方法检测正常成年豚鼠内耳中能进行细胞再生的位点。结果:正常成年豚鼠内耳中PCNA阳性产的定位于血管纹上皮细胞,螺旋缘齿间细胞,椭圆中分上皮细胞和内淋巴囊上皮细胞的胞核内,而耳蜗内,外毛细胞PCNA免疫反应阴性。结论:上述PCNA免疫反应阳性细胞是内耳中能进行细胞增殖的位点,成年哺乳动物耳蜗毛细胞不能进行细胞再生。

Abstract:Objective To investigate the proliferating cells in the inner ear of adult mammal.Methods Monoclonal antibody against proliferating cell nuclear antigen(PCNA)was used to identify the site of the cell renewing system in normal adult guinea pig by immunohistochemical methods.Results The PCNA positive products located in the nuclei of epithelial cells of strial vascularis, interdental cells of spiral prominerce,part of epithelial cells of utricle and epithelial cells of endolymphatic sac, whereas the positive products were not found in the hair cells of cochlea.Conclusion The cells labeled with anti PCNA antibodies which mentioned above are the sites of proliferation in the inner ear,and the negative result in the hair cells of cochlea provides further evidence that hair cells of cochlea in adult mammal can not be regeneration.
