养血清脑颗粒对肾性高血压大鼠血压及血管活性物质的影响

摘    要:目的探讨养血清脑颗粒对肾性高血压大鼠(Renovascular hypertension rat,RHR)的降压作用及对内皮素(Endothelin,ET)、一氧化氮(Nitric Oxide,NO)、降钙素基因相关肽(Calcitonin gene related peptide,CGRP)等的影响.方法采用改进的Goldblatt肾性高血压大鼠模型.造模成功后大鼠共分6组:模型组、养血清脑颗粒高、中、低3个剂量组、卡托普利组、牛黄降压丸组,每组10只,每日灌胃1次,正常组和假手术组灌服等量蒸馏水.分别在第1、2、4周的周五、六、日测量大鼠尾动脉收缩压.4周后取血,检测血中ET、NO、CGRP的浓度.结果 RHR模型大鼠血浆中ET含量增多,NO、CGRP含量降低,养血清脑颗粒各剂量降低血压同时能够降低ET含量,增加NO、CGRP含量,以高剂量组为优(P<0.05).结论养血清脑颗粒能够降低RHR血压,并对ET、NO、CGRP等产生一定影响,这可能是降压作用的机制之一.

Abstract:Objective To evaluate the effects of Yangxue Qingnao Granules (YQG) in decreasing blood pressure in rats with renal hypertension and to investigate its mechanism.Methods Modified Goldblatt renal hypertension rat models were used.The rats were randomized into 6 groups: model group,YQG groups (treated with high-,moderate-and low-dose respectively),Captopril Tablets group and Niuhuang Jiangya Pills group, 10 rats in each group. The model group was given with distilled water and the other groups with corresponding drugs by gavage, once time a day. Blood pressure was detected in 1st, 2nd and 4th week after treatment. Four weeks after treatment plasma levels of endothelin (ET),calcitonin gene related peptide(CGRP) and nitric oxide(NO) were examined by radioimmunoassay.Results Plasma level of ET was increased and those of NO and CGRP were decreased in rats with renal hypertension. YQG could decrease ET level and increase the levels of NO and CGRP, high-dose YQG in particular (P < 0.05).The blood pressure was decreased after one-month treatment of YQG. Conclusion YQG has an effect in decreasing blood pressure of rats with renal hypertension and its machanism may be related to the regulation of plasma levels of ET, CGRP and NO.
