高脂血症性脂肪性肝病大鼠肝脏Intermedin的表达

摘    要:目的研究高脂血症性脂肪性肝病大鼠血浆及肝组织intermedin(IMD)的变化,探讨新的小分子活性肽IMD在高脂血症性脂肪肝大鼠病理过程中的意义。方法雄性SD大鼠16只,随机均分为正常组与高脂组。高脂饲料喂养8周后,观察大鼠肝脏病理学变化,检测大鼠血脂及肝功能;放免法测定血浆及肝组织匀浆IMD含量。结果①高脂组大鼠体重、肝脏湿重和肝指数均显著高于正常组(P均〈0.01)。②高脂组TC、TG、LDL-C、ALT及AKP均显著高于正常组(P均〈0.01),HDL-C则低于正常组(P〈0.05)。③高脂组肝组织匀浆IMD水平较正常组高51.3%(P〈0.01),血浆IMD水平两组间差异无统计学意义(P〉0.05)。结论IMD可能在脂质代谢紊乱对肝损害的过程中扮演了重要的角色。

Abstract:Objective To investigate the changes of intermedin (IMD) in plasma and hepatic tissues of rats with hy- perlipoidemia and to study the possible role of IMD in the development of fatty Liver. Methods Sixteen male Sprague- Dawley rats were randomly divided into control group and hyperlipoidemia treatment group (each n = 8). The hyperli- poidemia models in rats were established by feeding with high-fat diet for 8 weeks. Liver histology, liver index (liver weight/body weight) , liver function ( ALT and AKP) and blood lipid were assayed. The contents of IMD in plasma and liver were measured with radioimmunoassay. Results The body weight, liver weight and liver index were increased strikingly in hyperlipoidemia treatment group compared with that in control group (P 〈 0.01 ). Liver histology showed mild to moderate fatty denaturation in hyperlipoidemia treatment group. TC, TG, LDL-C, ALT and AKP of hyperlipoi- demia experiment group were higher than those of control group ( respectively,P 〈 0.01 ) ,while HDL-C was lower than that of control group (P 〈 0.05). The IMD level in the hepatic tissues was increased by 51.3% in hyperlipoidemia treatment group compared with that in control group (P 〈 0.01 ) , while the plasma concentration was no difference between the groups (P 〉 0.05). Conclusion IMD may be correlated with the hyperlipoidemia and fatty liver in rats chro- nically fed with high fat diet.
