恩度对人胃癌裸鼠移植瘤的抑制效应及对Bcl-2、Bax基因表达的影响

 目的 探讨恩度对人胃癌裸鼠移植瘤的抑制效应及机制。方法 用体外培养的SGC-7901人胃癌细胞细胞制作荷瘤裸鼠，研究恩度作用对肿瘤生长的抑制效应。分别采用实时荧光定量PCR法、免疫组织化学染色法研究肿瘤组织中Bax及Bcl-2在mRNA及蛋白水平表达的变化。结果 恩度治疗组肿瘤体积明显小于对照组（P<0.05），实验第7、10、13、16、19天恩度的抑瘤率分别为26.2%、47.7%、38.9%、45.8%、51.7%。实时荧光定量PCR法结果表明，恩度组作用后Bcl-2在mRNA水平的表达显著低于对照组（P<0.05），但对Bax的表达无影响（P>0.05）。免疫组织化学结果与实时荧光定量PCR法结果一致。结论 恩度可显著抑制裸鼠SGC-7901人胃癌的生长，可能是通过降低Bcl-2的表达而发挥其杀伤效应。

The inhibitory effect of endostatin on human gastric carcinoma cells in an athymic mouse model and the impact on expression levels of Bcl-2 and Bax

Objective To investigate the inhibitory effect and the mechanism of endostatin on human gastric carcinoma model in nude mice. Methods Athymic mice were injected by in vitro-cultured SGC-7901 human carcinoma cell strain to observe the inhibitory effect of endostatin.
