白藜芦醇对人喉鳞癌细胞增殖的影响及其机制探讨

摘    要:目的观察白藜芦醇对喉鳞癌细胞增殖的影响,并探讨其机制。方法用不同浓度(12.5、25、50、100、200、400μmol/L)的白藜芦醇处理人喉鳞癌Hep-2细胞24、48、72 h,为白藜芦醇组。对照组不加白藜芦醇。采用MTT法测算Hep-2细胞增殖抑制率,采用TRAP-ELISA法检测Hep-2细胞端粒酶活性,采用W estern b lot法检测Hep-2细胞中的人端粒酶逆转录酶(hTERT)蛋白。结果白藜芦醇组Hep-2细胞增殖明显受到抑制,且呈浓度依赖性;细胞端粒酶活性受到抑制,hTERT蛋白水平明显下调,且呈浓度和时间依赖性。结论白藜芦醇可抑制喉鳞癌细胞增殖,与其下调癌细胞hTERT表达、抑制端粒酶活性有关。

Abstract:Objective To investigate the effect and mechanism on human laryngeal carcinoma Hep-2 cell proliferation. Methods Human laryngeal cancer Hep-2 cell in resveratrol group were treated with 12.5,25,50,100,200,400μmol/L resveratrol for 24,48,72 h. Hep-2 cell proliferation ratio was detected with MTT assay, and telomerase activity was detected by PCR-ELISA, hTERT protein was determined by Western blot,respectively. Results Hep-2 cell proliferation ratio , telomerase activity and hTERT protein expression in resveratrol group down-regulated in dose- and time-dependent manneres, respectively. Conclusion Resveratrole may suppress human laryngeal cancer Hep-2 cell proliferation through inhibiting telomerase activity and hTERT protein expression.
