摘    要:目的 构建弓形虫新基因wx2黄色荧光蛋白标记的黄色荧光超表达质粒,为进一步研究弓形虫新基因WX2的功能提供工具.方法 采用PCR扩增出编码wx2目的 基因,用BglⅡ,AvrⅡ分别对PCR扩增产物和ptubYFP-YFP/sagCAT进行双酶切.将wx2定向克隆到tubYFP-YFP/sag-CAT;对重组质粒进行PCR,双酶切初步鉴定后做序列测定.结果 特异扩增出预计wx2目的 基因片段,大小为558 bp;扩增产物成功连接到ptubYFP-YFP/sagCAT中,经PCR,双酶切及序列测定结果表明重组质粒中含有基因wx2读框.结论 成功构建弓形虫新基因wx2黄色荧光超表达质粒wx2一tub-YFP sag/CAT.
