摘    要:目的:观察人类野生型p53(wtp53)基因对人胰腺癌细胞的抑制作用。方法:用逆转录病毒为载体将外源性wtp53基因导入人胰腺癌细胞系PC-2,通过体外及小鼠体内实验研究转入基因的表达及对肿瘤的抑制作用,结果:p53在转染细胞PC-2/p53中表达水平提高,外源性wtp53基因的导入和表达能使PC-2细胞的生长速率减低,软琼脂集落形成能力下降,G0+G1期细胞比例增加,凋亡指数升高,裸鼠体内成瘤能力明显下降,结论:逆转录病毒载体介导的外源性野生型P53能抑制人胰腺癌细胞生长。

Abstract:Objective To observe the inhibitory effects of human wild-type p53(wtp53) gene on human pancreatic cancer cell line. Methods Retroviral vector was used to introduce exogenous wtp53 gene into human pancreatic cancer cell line PC-2, Gene expression and tumor inhibition were studied in vitro and in vivo . Results The expression of p53 in transfected cell lines(PC-2/p53) increased. The introduction and expression of exogenous wtp53 gene resulted in decreased growth rate and reduced ability to form colony in soft agar in PC-2 cells. The G 0+G 1 ratio increased and S ratio decreased in cell cycle distribution, and apoptosis index significantly increased in PC-2/p53 cells. The tumorigenicity of PC-2/p53 cells in nude mice was significantly suppressed by exogenous p53. Conclusion Exogenous wtp53 mediated by retroviral vector can inhibit the growth of human pancreatic cancer cell.
