摘 要: | 目的探讨结核分枝杆菌km基因突变的情况。方法通过传统梯度药敏试验和聚合酶链反应-单链构象多态性(PCR-SSCP)分析248株分枝杆菌临床分离株耐药情况和km基因的变化。结果药敏试验耐药率为32.7%(81/248),248株分枝杆菌临床分离株的16S rDNA SSCP电泳图谱均与结核分枝杆菌标准株相同。81株耐卡那霉素分离株中,24株(29.6%)km基因SSCP泳动异常。结论其部分耐药原因可能是由于T.M耐KM药物的km基因突变所致。

Abstract: | Objective To study the relationship between Km gene mutation and the drug-resistant M.tuberculosis.Methods Kanamycin drug-resistance and Km gene mutation in 248 M.tuberculosis strains isolated from clinical sputum specimens were analyzed by traditional drug susceptibility tests and PCR-SSCP.Results The rate of drug susceptibility tests was 32.7%(81/248).The electrophoresis profiles of 16S rDNA PCR-SSCP indicated that the 248 M.tuberculosis strains were according to M.TB standard strain.24 of the 81 kanamycin drug-resistant strains(29.6%) displayed abnormal in PCR-SSCP profiles.Conclusion In some degree,Km gene mutation made resistance to kanamycin of M.tuberculosis strains. |
