Egr-1诱骗性寡脱氧核苷酸对大鼠动脉损伤后bFGF表达的影响

摘    要:目的观察Egr-1诱骗性寡脱氧核苷酸(decoy ODN)对碱性成纤维细胞生长因子(bFGF)表达的影响。方法RT-PCR法测定血管壁组织的bFGF表达,采用Western-blot方法测定血管组织bFGF蛋白含量,以观察Egr-1诱骗性脱氧核苷酸对大鼠颈总动脉损伤后bFGF表达的影响。结果decoy ODN作用后,bFGFmRNA各时间点蛋白条带灰度减弱,bFGFmRNA表达减少。结论decoyODN可能是通过特异性抑制bFGF的表达,达到减轻新生内膜厚度,预防再狭窄的目的。

Effects of decoy oligodeoxynucleotide Egr-1 mRNA on the expression bFGF after balloon injury of rat common carotid artery

Abstract:Objective To observe the effects of the decoy oligodeoxynucleotide(ODN) Egr-1(early growth response factor-1) on bFGF expression after artery balloon injury,and to explore its possible mechanism of inhibiting neointimal formation.Methods Ninety six male Wister rats were randomly divided into four groups.Endothelial denudation of rats was performed with 2F balloon catheter except rats in the sham groups.Decoy ODN was transfected into the artery wall of the decoy ODN group by FuGENE6 during operation.The sham group,the simple injury group and the scrambled decoy ODN group servedas control groups.Six rats of each group were killed on the 7th,14th,21th day after balloon injury.The expressions of bfGF were performed by RT-PCR,Western-blot and immunohistochemical methods.Results(1) The level of bFGF mRNA began to increase on the 7th day and got to the peak on the 14th day.The positive expression of bFGF,protein was obvious on the 3rd day.The expression of bFGF reached its peak on the day 14 and begun to decrease on the day 14.(2) Compared with two groups,the expression level of bFGF mRNA and the expression of bFGF protein in FuGENE6 decoy ODN-group reduced at every time point(P<0.001).Conclusions The level of bFGF mRNA are increased in the process of intimal thickening,decoy ODN can decrease the expression level of bFGF mRNA after artery balloon injury.
