| 摘 要： | 目的探讨肺组织中基质金属蛋白酶(MMP-2/9)和其抑制物基质金属蛋白酶组织抑制因子(TIMP-1/2)在SiO2致矽肺纤维化中的表达及意义。方法清洁级Wistar大鼠64只,随机分为对照组和染尘组。采用暴露式气管内注入法染尘,染尘组每只大鼠注入1 ml SiO2混悬液,对照组注入1 ml灭菌生理盐水。染尘后3,7,14和28 d处死取其肺组织,光镜观察其病理切片;通过ELISA法,测定大鼠肺组织中MMP-2、MMP-9和TIMP1、TIMP-2蛋白的表达。结果与正常对照组比较,染尘组大鼠染毒后3 d肺组织发生局灶性病变,肺泡间隔增厚,间质细胞明显肿胀,局部可见肺泡腔形成细胞性结节,28 d时染尘组大鼠肺泡结构破坏或消失,肺组织以胶原沉积、肺纤维化改变为主,可见巨大纤维性结节;染尘组肺组织内MMP-2,MMP-9,TIMP-1和TIMP-2表达均较对照组增高,结果经秩和检验差异具有统计学意义(P<0.05)。结论矽肺组织内MMP-2/9和TIMP-1/2的高表达,说明其参与了矽肺纤维化的形成。
