两性霉素B合并氟康唑对体外培养新生隐球菌酚氧化酶的影响

摘    要:目的:研究两性霉素B合并氟康唑对体外培养新生隐球菌酚氧化酶活性的影响。方法:Novozyme234酶消化法获取原生质体;冷冻玻璃珠粉碎原生质体;Polecheck法测定酚氧化酶活性。结果:两性霉素B合并氟康唑组的新生隐球菌酚氧化酶活性较空白对照组明显降低(P<0.001),但在大于和接近最低抑菌浓度上限的组间无明显差异。结论:两性霉素B合并氟康唑可降低新生隐球菌酚氧化酶活性,在大于或接近最低抑菌浓度上限时与药物浓度无关。

Effect of amphotericin B united with fiuconazole on Cryptococcus neoformans phenoloxidases activity
Abstract:AIM: To study the effect of amphotericin united with fluconazole on Cryptococcus neoformans phenoloxidase activity. METHODS: Protoplast was acquired by Novozyme 234 enzyems digestion. Protoplast was broken by glass beads in freezing condition. Phenoloxidase activity was detected as Polecheck. RESULTS: The phenoloxidase activity of amphotericin B and fluconazole group was lower than control group ( P <0 001). But there wasn't significant difference when amphotericin B and fluconazole concentrations reached or exceeded the MICs. CONCLUSION: Amphotericin B united with fluconazole can decrease the phenoloxidase activity of cryptococccus neoformans . When the concentrations are near or higher than MICs, the activities have nothing to do with durgs concentrations.
