大鼠阿霉素慢性心衰模型的制备与心衰指标的判定

摘    要:目的探讨大鼠阿霉素(adrinmycin,ADR)慢性心衰(chronicheartfailure,CHF)模型的制备与心衰指标的判定。方法采用ADR给大鼠隔日腹腔注射制作慢性心衰模型,并通过行为体征观察、初体重和终体重、心重及心体比、心功能测定、心、肝和肺病理学光镜以及心肌病理学电镜观察进行心衰指标的判定。结果模型组大鼠萎靡不振、活动、进食减少、呼吸加快、体温下降、体重明显减轻,且左室内压、压力升高最大速率(±dp/dtmax)、心率明显降低,心脏病理学改变明显,基本符合Bishop关于慢性心衰CHF动物模型制作的标准,说明用ADR方法复制CHF模型是成功的。更贴近临床心肌病性CHF的病理生理过程。结论用阿霉素制作CHF模型技术方法容易控制,心衰指标符合标准。

Abstract:Objective To establish a chronic heart failure(CHF) model in rats by adrinmycin(ADR) and to study the indexes of judgment. Methods The model of CHF by the intraperitoneal injection(ip) of ADR once every two days was established, The indexes of CHF through the observation of behavior, body weight, heart weight, ratio of heart weight and body weight, detection of heart function, observation of pathological changes in liver and lung under the light microscope as well as the myocardial pathological changes under the electroscope were studied. Results Model groups rats showed inanimate behavior, decrease of the body temperature, activities and food intake, faster breathing, significant loss of weight, increasing pressure of left ventricular, the significant decrease of heart rate. the myocardial pathological changes were obvious, too. The model basically hits the target of Bishop indexes of CHF which suggests that the CHF model of rats by ADP was successful one that was more similar with the pathological processes of cardiomyopathy style CHF. Conclusion Model of CHF induced by ADR is reliable and easily both copyed and controled.
