摘    要:急性白血病的出凝血异常和组织因子(tissue factor,TF)的高表达有关.循环血流中的TF主要表达于细胞、微粒(microparticle,MP)和异常剪接TF(altematively spliced human tissue factor,asHTF)中.为探究asHTF在急性髓系白血病(acute myeloblastic leukemia,AML.)出凝血异常中的作用,本研究选用了6种常见的AML细胞株NB4、BL-60、Kasumi-1、U937、K562和THP-1,在RNA水平进行了RT-PCR检测.结果发现,在6种细胞株中仅NB4、U937细胞存在全长TF和asHTF的RNA水平基线表达并经测序验证,对上述2种细胞在蛋白质水平进行了流式细胞仪及TF活性、杭原检测发现,asHTF仅有极少量TF抗原表达且基本无TF活性,而MP相关TF(microparticle associated tissue factor,MP-TF)的抗原和活性显著高于asHTF.结论:在NB4、U937细胞中asHTF基本无促凝作用,而MP-TF有凝血活性且在肿瘤细胞释放TF中占主导地位.
