| 摘 要： | 目的研究骨转移瘤原代细胞中是否存在干细胞样侧群细胞(SP细胞)。方法对乳腺癌、非小细胞肺癌、前列腺癌、肾癌及宫颈癌骨转移瘤原代细胞进行Hoechst33342染色,应用荧光显微镜直接进行观察,检测骨转移瘤原代细胞中是否存在SP细胞及非SP细胞,并给予维拉帕米拮抗剂,观察各骨转移瘤瘤细胞中SP细胞亚群染色变化情况。结果所检测的乳腺癌、非小细胞肺癌、前列腺癌、肾癌及宫颈癌骨转移瘤原代细胞中均存在SP细胞。荧光显微镜下,SP细胞体积较非SP细胞小,SP细胞胞核蓝色荧光呈淡染或消失;非SP细胞经染色后发出较强的蓝色荧光。给予维拉帕米拮抗剂后,各骨转移瘤细胞中胞核发蓝色荧光的细胞数目明显增多。结论乳腺癌、非小细胞肺癌、前列腺癌、肾癌及宫颈癌骨转移瘤原代细胞中存在干细胞样的SP细胞。 |
