氧化修饰低密度脂蛋白的蛋白组份可引起巨噬细胞线粒体膜的损伤

摘    要:采用ACAS570测定小鼠腹腔巨噬细胞线粒体膜电位,观察了氧化修饰低密度脂蛋白(Ox-LDL)对巨噬细胞线粒体膜的损伤及其不同组份(moiety)在致损伤中的作用。结果显示Ox-LDL可使巨噬细胞线位体膜产生损伤,用GSHPx模拟物ebselen清除Ox-LDL上的脂氢过氧化物(LOOH)可使损伤减轻20%。单独的脂质组份虽然含有大量的LOOH,但对巨噬细胞线粒体膜电位无显著影响,而单独的蛋白组份则可引起线粒体膜电位下降,其降低程度与Ox-LDL清除LOOH后的降低程度相当,说明Ox-LDL清除LOOH后对线粒体膜的损伤可能是由修饰的载脂蛋白B(apoB)产生的。

Abstract:The role of lipid hydroperoxides(LOOH),the lipid and protein moiety in Ox-LDL in the injuryto macrophage mitochonodrial membrane induced by Ox-LDL was observed by measuring mitochondrialmernbrane potential on ACAS570.The results showed that the mitochondrial membrane potential de-creased when macrophages were treated by Ox-LDL,If LOOH in Ox-LDL was precleared by ebselen plusGSH,the decreased mitochondrial membrane potential could be recovered by about 20%. The lipid moietyalone although containing a large amount of LOOH had no significant effect on mitochondrial membranepotential.The protein moiety alone could make the membrane potential decrease,and the degree of the de-crease was equivalent with that of Ox-LDL in which LOOH was cleared by ebselen plus GSH.
