摘    要:目的:探讨E2F-1对人胃癌细胞MGC-803生物学行为的影响。方法:利用脂质体将pCMV-E2F-1-HA质粒和pCMV-HA质粒分别转染人胃癌细胞MGC-803,应用RT-PCR和蛋白质印迹法检测MGC-803细胞中E2F-1基因的表达;应用体外侵袭实验、迁移实验、细胞基质黏附实验和克隆实验分别检测E2F-1对胃癌细胞MGC-803的侵袭力、迁移力、黏附能力和增殖力的影响。结果:转染pCMV-E2F-1-HA质粒的MGC-803细胞检测到E2F-1的表达;转染pCMV-E2F-1-HA质粒的MGC-803细胞的侵袭力、迁移能力和增殖力较转染pCMV-HA质粒的MGC-803细胞和未转染的MGC-803细胞明显降低(P<0.05),细胞黏附能力未见明显变化,P=0.072。结论:E2F-1高表达对人胃癌细胞MGC-803的生物学特性有较大的影响,具有抑制其侵袭转移的作用。

Abstract:OBJECTIVE:To explore the effects of E2F-1 on behaviors of human gastric carcinoma cell line MGC-803.METHODS:An eukaryotic expression vector was constructed to transfect a human gastric carcinoma cell line MGC-803.In MGC-803 cells transfected by pCMV-E2F-1-HA,E2F-1 gene was detected by RT-PCR and Western blot techniques.The cell matrigel invasion assay,adhesion assay,migration assay,and cloning assay were used to study the effects of E2F-1 gene.RESULTS:E2F-1 mRNA and protein were detected in MGC-803 cells transfected by pCMV-E2F-1-HA.The invasiveness,migration and proliferation were inhibited significantly in MGC-803 cells transfected by pCMV-E2F-1-HA compared with that in MGC-803 cells transfected by pCMV-HA and untransfected cells,P<0.05.The adhesive ability of MGC-803 cells with the matrix was not affected by the transfection,P=0.072.CONCLUSIONS:E2F-1 can affect the behaviors of human gastric carcinoma cell line MGC-803.It suggests that E2F-1 may depress the invasion and metastasis of gastric carcinoma cells.
