摘    要:目的探讨姜黄素对人胆囊癌细胞株QBC939增殖及凋亡的影响,为胆囊癌的临床治疗提供新的思路。方法姜黄素与QBC939共同孵育后,采用MTT方法测定QBC939的增殖情况并计算抑制率,用流式细胞仪评估凋亡率,同时以蛋白免疫印迹(Western blot)方法检测姜黄素对凋亡相关蛋白Caspase 3表达水平的影响。数据比较采用t检验,P<0.05为差异有统计学意义。结果姜黄素处理组细胞生长明显受抑并存在显著凋亡,Western blot结果显示Cas-pase 3表达量远大于对照组。结论姜黄素可显著抑制胆囊癌细胞株QBC939的增殖,这种变化可能与其诱导的Cas-pase 3的高表达及促进凋亡有关。

Abstract:Objective To evaluate the impacts of curcumin(Cur) on proliferation and apoptosis of gallbladder carcinoma cell line QBC939 to provide the new ideas for its clinical treatment.Methods After coincubation of QBC939 with Cur solution,the MTT assay was utilized to detect the proliferation circumstance of QBC939 cells and the inhibiting rate was calculated.Flow cytometry was used to determine the apoptosis rate of QBC939 cells.Meanwhile,the apoptosis-related protein Caspase-3 was measured via Western blot.All data were analyzed using t test and the statistical difference was considered when the value of P was lower than 0.01.Results In the Cur group,the cell proliferation was significantly inhibited and apoptosis presented evidently.The Western blot results revealed that the expression amount of protein Caspase 3 in the Cur group was bigger than that in the control group.Conclusion Curcumin can significantly suppress the proliferation of gallbladder cancer cell line QBC 939,this change may be related with the curcumin-inducing high expression of Caspase 3 and promoting apoptosis.
