哮喘患者外周血CD4+CD25+调节性T细胞与IL- 4,IL- 5的表达

摘    要:目的:探讨哮喘患者外周血中CD4 CD25 调节性T细胞与IL- 4,IL- 5的水平变化及其相互关系.方法:采用流式细胞技术检测40例哮喘患者和40例健康对照者外周血中CD4 CD25 调节性T细胞的表达水平,采用ELISA法检测外周血中IL- 4,IL- 5水平.结果:哮喘患者外周血中CD4 CD25 调节性T细胞低于健康对照者(P<0.01);急性发作期哮喘患者外周血中CD4 CD25 调节性T细胞低于非急性发作期(P<0.01);哮喘患者IL- 4,IL- 5的表达水平高于健康对照者(P<0.05);急性发作期患者IL- 4,IL- 5的表达水平高于非急性发作期(P<0.05);急性发作期哮喘患者外周血CD4 CD25 T细胞与IL- 4,IL- 5的表达水平呈负相关(r分别为-0.675,-0.767,P<0.01).结论:CD4 CD25 调节性T细胞在哮喘的疾病活动中起着重要作用,是导致哮喘患者细胞免疫失调的重要原因.

Abstract:AIM: To evaluate the changes and the correlation between regulatory CD4+CD25+ T cells and IL- 4,IL- 5 in peripheral blood from the patients with asthma. METHODS: Forty patients with asthma and 40 healthy volunteers were included in this study. The proportion of CD4+CD25+ T cell population in total T cells was evaluated by flow cytometric analysis.The serum IL- 4 and IL- 5 levels were measured by ELISA. RESULTS: The expression of CD4+CD25+ T cells in peripheral blood from the patients with asthma was much lower in comparison with the healthy volunteers(P<0.01) ; The expression of CD4+CD25+ T cells in peripheral blood from the patients during acute attacks was lower than that in chronic phase(P<0.01); IL- 4, IL- 5 levels from the patients with asthma were higher than those in healthy volunteers(P<0.05); IL- 4, IL- 5 levels during acute attacks were higher than those in chronic phase(P<0.05); The expression of CD4+CD25+ T cells in peripheral blood was in negative correlation with the levels of IL- 4, IL- 5 from the patients with asthma during acute attacks (r=-0.675, -0.767, P<0.01). CONCLUSION: Regulatory CD4+CD25+ T cells play a very significant role in activation of asthma, and is the important reason for the maladjustment of cellular immunofunction in the patients with asthma.
