党参总皂苷对大鼠局灶性脑缺血性损伤的保护作用

摘    要:目的观察党参总皂苷(TSC)对大鼠局灶性脑缺血性损伤的保护作用,并初探其机制。方法 54只SD大鼠随机分为假手术组、假手术给药组、模型组、治疗组。假手术给药组及治疗组每日1次TSC60mg·kg~(-1)灌胃,假手术组及模型组灌胃等体积的生理盐水,7d后建立大鼠右侧大脑中动脉阻塞(MCAO)的局灶性脑缺血模型,24h后观察各组大鼠死亡率、神经功能症状,2,3,5-三苯基氯化四氮唑(TTC)染色测量脑梗死体积,real time RT-PCR法测定缺血周边区肿瘤生长因子α(Tnf-α)、白细胞介素1β(Il-1β)、环氧酶2(Cox-2)、内皮型一氧化氨合酶(Nos3)及诱导型一氧化氮合酶(Nos2)的mRNA表达。结果 TSC明显降低MCAO所致的死亡率及神经功能评分,并减少脑梗死体积(均P<0.05)。同时,TSC明显抑制了缺血周边区Tnf-α、Il-1β、Cox-2、Nos3及Nos2的mRNA表达(P<0.05),而对假手术给药组大鼠无明显影响(P>0.05)。结论 TSC对大鼠局灶性脑缺血性损伤有保护作用,其机制与抑制炎症相关基因的mRNA表达有关。

Abstract:AIM To investigate the protective effects of total saponin of Radix Codonopsis(TSC) on focal cerebral ischemia in rats,and explore the possible mechanisms of action.METHODS Fifty-four Sprague Dawley rats were randomly divided into sham group,sham-treated group,model group and model-treated group. Rats in sham-treated and model-treated groups were administered by TSC at the dose of 60 mg·kg~(-1) by gavage, while sham and model groups were administered by the same volume of normal saline instead.Seven days later, the focal cerebral ischemia model was induced by right middle cerebral artery occlusion in rats.Twenty-four hours after the operation,mortality and the neurological function in each groups were observed,and cerebral infarction volume was determined by 2,3,5-triphenyltetrazolium chloride(TTC) staining.For the mechanism study,the mRNA expressions of Tnf-α,Il-1β,Cox-2,Nos3 and Nos2 in ischemic border were detected by real time RT-PCR.RESULTS TSC at dose of 60 mg·kg~(-1) caused a significant decrease in mortality,the score of neurological function and cerebral infarction volume(P<0.05).TSC inhibited the mRNA expressions of Tnf-α, Il-1β,Cox-2,Nos3 and Nos2 in ischemic border(P<0.05),while it did not affect those of the sham-treated group(P>0.05).CONCLUSION TSC has protective effects on focal cerebral ischemia in rats,which may be related to the inhibition of mRNA expressions of inflammation-related genes.
