龙葵总碱对骨髓瘤细胞凋亡及NF-KB表达的影响

摘    要:目的:探讨龙葵总碱对人骨髓瘤RPMI-8226细胞的凋亡作用以及对NF-KB表达的影响。方法:选用人骨髓瘤RPMI-8226细胞株为研究对象,以龙葵总碱为被试对象,采用MTT法检测细胞增殖活性,AnnexinV-FITC/PI双标记法分析细胞凋亡的改变;免疫荧光细胞化学染色分析NF-KB的表达情况。结果:在25mg/L龙葵总碱处理RPMI-8226细胞48h后,RPMI-8226细胞的生长出现明显的抑制作用,且随着浓度加大及时间延长,其抑制作用逐步增强;药物浓度〈25mg/L时,诱导凋亡作用不明显,当药物浓度达到25mg/L时,早期凋亡明显增多;当药物浓度达25mg/L(此时细胞凋亡明显)时,NF-KB表达水平的降低具有统计学意义。结论:龙葵总碱可抑制骨髓瘤RPMI-8226细胞增殖,促进骨髓瘤细胞凋亡,并且表现为剂量和时间依赖性;NF-KB表迭量在人多发性骨髓瘤RPMI-8226细胞株随龙葵总碱的药物浓度增大而降低。

Effects of coramsine on apoptosis and NF - KB expression of myeloma cells

Abstract:Objective : To investigate the effects of coramsine on the apoptosis and nuclear factor - kappa B ( NF - KB ) expression of human myeloma cells (RPMI -8226 ). Methods:Human myeloma cell line RPMI -8226, which was exposed to coramsine, was selectedas the subject. Cell viability was measured by MTT assay;cell apoptosis was analyzed by annexin V - FITC/PI double staining;the ex pression of NF - KB was measured by immunofluorescent cytochemical staining. Results : RPMI - 8226 cells showed significantly inhibi ted growth after being treated with 25 mg/L coramsine for 48 h, and the inhibitmy effect was enhanced as the concentration of coramsine increased and treatment proceeded. When the concentration of coramsine was less than 25 rag/L, the induced apoptosis of RPMI - 8226 cells was not notable;when the concentration reached 25 mg/L, early apoptosis increased significantly, and the expression of NF - KB decreased significantly. Conclusion : Coramsine can inhibit proliferation and promote apoptosis in myeloma cell line RPMI - 8226, which is dose - and time - dependent. The expression of NF - KB in human multiple myeloma cell line RPMI - 8226 decreases as the concen tration of coramsine increases.
