千金益康片对去卵巢模型大鼠E2、FSH及相关神经递质的影响

摘    要:目的:观察千金益康片对去卵巢模型大鼠血清雌二醇(E2)、卵泡刺激素(FSH)及相关神经递质的影响。方法:将60只生育龄雌性未孕SD大鼠随机分为空白对照组,模型对照组,千金益康片低、中、高剂量组和阳性药组,每组各10只,除空白对照组外,其余5组均切除卵巢;千金益康片低、中、高剂量组分别给予千金益康片混悬液5.3、10.6、21.18g/kg体质量灌胃,阳性药组给予女珍颗粒混悬液0.75g/kg体质量灌胃,空白对照组和模型对照组用等体积蒸馏水(10mL/kg体质量)灌胃,每天1次,连续14d;末次给药24h后测定各组大鼠E2、FSH水平,检测下丘脑神经递质5-羟色胺(5-HT)、去甲肾上腺素(NE)含量,计算子宫、肾上腺脏器系数。结果:与模型对照组比较,千金益康片中、高剂量组均能明显增加去卵巢模型大鼠血清E:水平(P〈0.05,P〈0.01);千金益康片低、中、高剂量组能明显降低去卵巢模型大鼠血清FSH水平(P〈0.05);低、中、高剂量组对下丘脑5-HT含量有降低趋势(P〈0.05);中剂量组能明显增加NE含量(P〈0.01),低、中、高剂量组能增加子宫系数(P〈0.05),而中、高剂量组能明显增加肾上腺系数(P〈0.05)。结论:千金益康片可增加去卵巢模型大鼠血清E2水平,降低血清FSH水平,增加NE含量,增加子宫系数和肾上腺系数,对5-HT有降低趋势,其作用机制可能与调节内分泌系统和神经系统有关。

Effects of Qianjin Yikang tablets on E2, FSH, and related neurotransmitters in ovariectomized rats

Abstract:Objective:To observe the effects of Qianjin Yikang tablets on estradiol( E2 ), follicle -stimulating hormone (FSH), and related neurotransmitters in ovariectomized rats. Methods : Sixty non - pregnant female Sprague - Dawley rats of reproductive age were randomly and equally divided into blank control group, model control group, low -, middle, and high -dose Qianjin Yikang tablet groups, and positive medicine group. The rats in model control group, Qianjin Yikang tablet groups, and positive medicine group were ovariectomized. The low - , middle - , and high - dose Qianjin Yikang tablet groups were given Qianjin Yikang tablets (5.3,10. 6, and 21.18 g/kg, respectively) by gavage. The positive medicine group received suspension of Nuzhen granules (0. 75 g/kg) by gavage. The blank control group and model control group received equal volumes of distilled water( 10 ml/kg). Medication was given once daily for 14 consecutive days. At 24 h after the last administration, serum E2 and FSH levels were measured, norepinephrine (NE) and 5 hydroxytryptamine( 5 -HT) in the hypothalamus were quantified, and the organ coefficients of uterus and adrenal gland were calculated. Results : Compared with the model control group, the middle - and high - dose Qianjin Yikang tablet groups had significantly increased serum E2 levels(P 〈0. 05 ,P 〈0. 01 ) ;the low - , middle - , and high -dose Qianjin Yikang tablet groups had significantly decreased serum FSH levels and showed a decreasing trend in 5 - HT content in the hypothalamus ( P 〈 0. 05 ) ; the middle - dose Qian jin Yikang tablet groups had significantly increased NE content (P 〈 0. 01 ) ;low -, middle - , and high - dose Qianjin Yikang tablet groups had increased organ coefficient of uterus, and the middle - , high -dose Qianjin Yikang tablet groups had significantly increased organ coefficient of adrenal gland. Conclusion:Qianjin Yikang tablets can decrease serum FSH level and increase serum E2 level, NE content, organ coefficient of uterus and organ coefficient of adrenal gland and tend to decrease 5 - HT content. Its action mechanism may be related to its regulatory effects on the endocrine system and nervous system.
