枳术饮对功能性消化不良大鼠VIP及其受体mRNA表达的影响

摘    要:目的:观察枳术饮对功能性消化不良(FD)大鼠血管活性肠肽(VIP)及其受体mRNA表达的影响,探讨该方促进肠道动力的作用机制。方法:将Wistar大鼠随机分为正常组、模型组、枳术饮组和西沙比利组。复制大鼠胃电节律失常模型,药物干预后,观察各组大鼠体重的变化。检测大鼠血浆及十二指肠组织VIP水平。观察十二指肠病理组织学改变。逆转录聚合酶链反应(RT-PCR)法测定大鼠十二指肠血管活性肠肽受体1(VIPR-1)的表达。结果:模型组大鼠体重明显减少(P0.01),血浆及十二指肠组织VIP含量明显增加(P0.01),十二指肠VIPR-1mRNA的表达明显增加(P0.01)。枳术饮可显著增加模型大鼠体重(P0.05),显著降低血浆及十二指肠组织VIP含量(P0.05),下调十二指肠VIPR-1mRNA的表达(P0.05)。结论:枳术饮能够降低FD大鼠血浆及十二指肠组织VIP水平,抑制FD大鼠十二指肠VIPR-1mRNA的表达,这可能是其促肠道动力的作用机制之一。

Abstract:Objective:To investigate the effect of ZhizhuYin on vasoactive intestinal peptide(VIP)and VIP receptor in rats with functional dyspepsia and its mechanism.Methods:Wistar rats were randomized into normal group,model group,Zhizhu Yin group and cisapride group.The rat model of electrogastric dysrhythmias was used.After drug intervention,the changes of rat weight had been observed.Changes in VIP contents in blood plasma and dodecadactylon tissue were determined.The changes of rat dodecadactylon pathohistology had been observed.The express of vasoactive intestinal peptide-receptor1(VIPR-1) in rat dodecadactylon was determined by RT-PCR.Results: Compared with that of the normal control group,the weight of FD rats was decreased significantly(P0.01)and the VIP level elevated significantly in blood plasma and dodecadactylon tissue(P0.01).The express of VIPR-1mRNA in rat dodecadactylon was increased significantly in model group(P0.01).Compared with that of the model group,Zhizhu Yin can upgrade the weight(P0.05)and degrade the VIP level(P0.05).It can the degrade express of VIPR-1mRNA(P0.05).Conclusion: Zhizhu Yin can degrade VIP in blood Plasma and dodecadactylon tissue.It can downregulate the express of VIPR1Mrna,which may be one of the mechanism in promoting intestinal motility.
