前列腺癌组织EZH2基因的表达及其意义

摘    要:目的:探讨EZH2基因在列腺癌组织中的表达意义. 方法:采用半定量RT-PCR检测前列腺癌、前列腺上皮内瘤(PIN),良性前列腺增生中EZH2表达状况并对其扩增产物进行测序. 结果:前列腺癌40例组织中,EZH2表达率(100%). 良性前列腺增生组织无表达, PIN 20例中仅有1例有表达. 晚期前列腺癌组织表达明显高于早期前列腺癌(P<0.05);低分化前列腺癌表达高于中高分化前列腺癌(P<0.05);激素依赖性前列腺癌表达高于激素非依赖性前列腺癌(P<0.05). EZH2表达高低与前列腺血清总PSA浓度无关(P>0.05). 结论:EZH2基因表达状况与前列腺癌分期、分级有关,其表达对判断前列腺癌病理生物学特性和前列腺癌治疗有重要意义.

Abstract:AIM: To determine the expression of EZH2 gene in prostate cancer(PCa) and to investigate the relation between EZH2 expression and biological characters of PCa.METHODS: In 40 cases of PCa,20 cases of benign prostate hyperplasia(BPH),20 cases of prostatic intraepithelial neoplasia(PIN),the expression of EZH2 was detected by semi-quantitative RT-PCR and the amplified product was sequenced.RESULTS: All 40 PCa cases were positive in EZH2 expression.There was no expression of EZH2 gene in BPH.Only one of the 20 PIN cases was seen EZH2 expressed positively.EZH2 expression was increased in the advanced compared with the primary PCa(P<(0.05)),in the low-differentiated compared with the highly-differentiated PCa(P<0.05) and in the hormone-dependent compared with the hormone-independent PCa(P<0.05).However,EZH2 expression did not have a linear correlation with the serum total prostate specific antigen level(P>0.05).CONCLUSION: EZH2 gene plays an important role in the development of prostaic carcinoma.Therefore,the analysis of EZH2 expression will benefit the diagnosis and treatment of prostatic carcinoma.
