聚合酶链反应对沙门氏菌肠炎快速诊断的初步应用

摘    要:对沙门氏菌肠炎快速诊断,采用聚合酶链反应和DNA序列分析,以沙门氏菌保守序列--侵袭基因A作为靶序列,检测23株沙门氏菌均扩增出284bp特异的DNA片段,提示存在侵袭基因A,20株非沙门氏菌扩增阴性。检测84份粪标本,13份阳性,其中仅8份大便培养阳性。DNA序列分析证实粪标本扩增出的284bpDNA片段与侵袭基因A序列一致。结果表明聚合酶链反应用于沙门氏菌肠炎诊断,具有简便、快速、敏感等优点,适于临床快速诊断

Abstract:The method of polymerase chain reaction(PCR)and DNA sequence analysis were used to detect salmonella enteroclitis quickly.As the target DNA sequence the invA gene of 284bp DNA segments were amplified in all 23 strains of salmonella by PCR and no any segments in 20 strains of non salmonella strains.84 feacil samples were amplified directily by PCR and 13 of them were positive and 8 of them were positive in culture.It was confirmed that DNA sequences of 284bp segments were consistent with the one of the invA gene.The results showed that PCR for salmonella is simple,quick and sensitive,which is suitabl to apply in clinic quick diagnosis of salmonella enterocolitis.
