| 摘 要： | 目的:考察缓释药物载体聚乳酸在脑内应用的安全性。方法:体外原代培养大鼠大脑皮层神经元,加入聚乳酸处理后通过细胞计数试剂盒(CCK-8)检测细胞毒性,Hoechst33258染色检测细胞凋亡,并与正常对照组比较;自制聚乳酸空白片,植入大鼠颅内30d后,经行为能力测试、HE染色和Nissl染色检测聚乳酸对脑神经元及其功能的影响,并与假手术组、正常对照组比较。结果:与正常对照组比较,聚乳酸组神经元培养至第7、9、12天时,CCK-8吸光度值及细胞凋亡率均无显著差异;30d后聚乳酸空白片组与假手术组比较,脑组织有轻微炎症反应,但神经元数量和形态功能没有发生显著变化。结论:聚乳酸对体外培养的大脑皮层神经元抑制作用和诱导凋亡作用不显著,对脑内神经组织无明显毒性。
