bFGF及其抗体对裸鼠皮下移植膀胱癌生长影响的研究

摘    要: 目的 探讨碱性成纤维细胞生长因子 (bFGF)及其抗体对裸鼠人膀胱癌移植瘤生长的影响和作用机制。方法 建立膀胱癌EJ细胞裸鼠皮下移植瘤动物模型 ,观察bFGF和bFGF抗体对裸鼠皮下瘤生长状况的影响 ;应用免疫组织化学方法检测肿瘤组织中增殖细胞核抗原 (PCNA)和Ⅷ因子相关抗原的表达。结果  (1)bFGF治疗组皮下瘤体积和重量分别比对照组显著增多 ,瘤组织中PCNA指数和微血管密度 (MVD)也显著提高 ;(2 )bFGF抗体治疗组皮下瘤体积和重量比对照组均显著减少 ,瘤组织中PC NA指数和MVD也显著降低。结论 bFGF能够促进膀胱癌生长而bFGF抗体具有抑制肿瘤的作用 ,其作用机制与调控肿瘤细胞增殖和血管形成有关

Abstract:Objective To investigate the effects of basic fibroblast growth factor (bFGF) and its antibody on xenograft bladder tumor in nude mice and elucidate their mechanisms.Methods The animal model of xenografted bladder cancer of EJ cell line was established in 40 nude mice and was randomly divided into following groups: bFGF group and control group, anti-bFGF antibody group and control group. The expressions of PCNA and microvessel density (MVD) in tumor tissue were assayed by immunohistochemistry.Results (1) The volume and weight of tumor, PCNA index and MVD of tumor tissue in bFGF treated group were significantly higher than those in control group,respectively;(2) The volume and weight of tumor, PCNA index and MVD of tumor tissue in anti-bFGF antibody treated group were significantly lower than those in control group,respectively.Conclusion bFGF can promote growth of bladder cancer while anti-bFGF antibody has antitumor effect on bladder cancer, and that at least in part, the important mechanisms of their action involve regulating bladder cancer cell growth and angiogenesis.
