血管生成素及受体和VEGF在人肝细胞癌组织中的表达及作用

目的 研究血管生成素及受体Tie-2、血管内皮生长因子(VEGF)在肝细胞癌血管生成和进展中的作用。方法 用RT-PCR和免疫组织化学方法分析了28例肝细胞癌、10例肝硬化和10例正常肝标本。探讨血管生成素及受体、VEGF的表达和HCC肿瘤血管生成和临床病理的关系。结果 Ang-2／Tie2、VEGF在肝细胞癌中表达明显上调，免疫组织化学也显示Ang-2和VEGF蛋白在肝癌表达增强。Ang-2／Ang-1mRNA比值与肿瘤血管侵犯及CD34染色的微血管密度相关。结论 Ang／Tie-2、VEGF在HCC肿瘤血管生成和进展中起重要作用。

Expression and significance of angiopioetins/Tie-2 and VEGF in hepatocellular carcinoma

Objective]To investigate the effects of Angiopioetins and tyrosine kinase receptor Tie-2, vascular endothelial growth factor (VEGF) on the angiogenesis, growth, and metastasis of hepatocellular carcinoma(HCC). With the methods of RT-PCR and immunohistochemistry. Specimens from 28 HCC patients, 10 cirrhotics, 10 normal livers were analysised. And the relationship between Angiopioetins, VEGF exprssion and the pathological characteristics of HCC was studied. Ang-2, Tie-2, VEGF menssenger RNA(mRNA) were significantly up-regulated in HCC compare to cirrhotic tissue and normal liver tissue. Immunohistochemical staining also showed increased Ang-2, VEGF protein in HCC. A high Ang-2/Ang-1mRNA ratio and high VEGF in HCC were closely associated with tumor vascular invasion,tumor diameter, and microvessel density level as assesssded by CD34. Conclusion ]Ang /Tie-2andVEGF may play a critical role in promoting tumor angiogenesis and progession in human HCC.
