侧脑室内注射复方丹参注射液对大鼠血压和心率的影响

摘    要:背景复方丹参注射液具有增加心肌耗氧量,降低血压和心率的作用,丹参注射液对心血管系统的作用机制可能不是经胆碱能系统起作用的.目的探讨复方丹参注射液降低大鼠动脉血压和心率的作用及机制.设计完全随机设计. 地点和对象实验地点郧阳医学院.实验用 SD雄性大鼠78只,清洁级,体质量 270~ 350 g.将大鼠随机分为 8组,即生理盐水组( n=8); 侧脑室内注射丹参组( n=10) ; 静脉注射丹参组( n=6) ;量效曲线组( n=16);生理盐水+丹参组( n=6) ;心得安+丹参组( n=11);酚妥拉明+丹参组( n=11);阿托品+丹参组(n=10). 干预在氨基甲酸乙酯麻醉、三碘季胺酚制动、呼吸机控制呼吸的SD大鼠上观察复方丹参注射液对大鼠血压和心率的影响,连续观察 60min血压和心率曲线变化,与给药前基础值比较,计算各观察指标在给药后不同时间的变化百分率,并与给药前的水平及生理盐水+丹参组比较.主要观察指标各组大鼠注药前后对血压和心率的变化. 结果侧脑室内注射丹参组一般在给药后30 s之内血压开始下降, 15 min时达最低值,由给药前的( 12.6± 1.5) kPa降至(9.7± 1.7) kPa,给药后 5, 10, 15 min与给药前相比差异均有显著性意义 ;给药后心率也下降,由给药前的(419.2± 18.5)次 /min下降到( 326.6± 14.3)次 /min,给药后 5, 10, 15 min与给药前相比,差异均有显著性意义(P< 0.01).生理盐水+丹参组血压、心率明显降低.心得安+丹参组观察到给予丹参注射液血压心率降低效应明显减弱.结论复方丹参注射液有明显降低大鼠血压和心率作用.侧脑室预先注射β-受体阻断剂心得安可部分阻断复方丹参注射液的降低血压和心率作用.

Abstract:BACKGROUND:Composite salvia miltiorrhiza(SM)injection can increase the myocardial oxygen consumption and reduce the blood pressure and heart rates.The effects of SM on cardiovascular system may not be caused by cholinergic system. OBJECTIVE: To explore the effects and mechanism of SM on blood pressure and heart rates of rats. DESIGN:Completely randomized design. SETTINGS and MATERIALS:Experimental site: Yunyang Medical College.Seventy-eight male SD rats of clearing grade,weighing 270 g to 350 g were chosen.Rats were randomly divided into 8 groups which was Saline group(n=8),lateral ventricle injection of SM group(n=10),intravenous injection of SM group(n=6), dose effect curve group(n=16), saline+ SM group(n=6), propanolol+ SM group(n=11),phentolamine+ SM group(n=11) and atropine+ SM group(n=10). INTERVENTIONS:The effects of SM on blood pressure and heart rates of 78 male SD rats were observed after using urethane as anesthesia,immobilizing of Gallamine and controlling respiration by respirator.The curve changes of blood pressure and heart rates were continuously observed for 60 minutes.The change percentage of each index after administration was calculated and compared with the foundational values before administration,as well as the saline+ SM group. MAIN OUTCOME MEASURES:Change of blood pressure and heart rates before and after administration of SM of rats in each group. RESULTS:In the lateral ventricle injection of SM group,the blood pressure started to reduce in the 30 s after administration and reached the minimal value in the 15 minutes which decreased from(12.6± 1.5) kPa before administration to(9.7± 1.7) kPa after administration.There was significant difference between pre-administration and 5, 10 and 15 minutes after administration.The heart rats also reduced after administration which decreased from( 419.2± 18.5) times/minute to( 326.6± 14.3) times/minute.There was also difference between pre-administration and 5,10 and 15 minutes after administration(P< 0.01).In saline+ SM group,the blood pressure and heart rates had greatly decreased.In propanolol+ SM group,it was found that the effects of SM on reducing blood pressure and heart rates were weakened. CONCLUSION:Composite salvia mitiorrhiza injection can greatly reduce the blood pressure and heart rates of rats.Injecting Propranolol,the βreceptor blocking agent through lateral ventricle in advance can partly block the effects of composite salvia mitiorrhiza injection on reducing blood pressure and heart rates.
