DAF与CD3协同刺激人外周血T细胞活化的实验研究

目的：探讨人促衰变加速因子（decay-accelerating factor,DAF)作为共刺激分子参与T细胞活化的作用机制。方法：观察3株针对DAF不同SCR的单抗单独使用或者与抗人CD3单抗联合使用时，对人外周血T细胞增殖、IL－2分泌以及胞浆Ca^2 水平的影响。结果：所用3株抗人DAF单抗不能活化T细胞，而抗人DAF单抗与抗人CD3单抗可以协同刺激T细胞增殖，促进IL－2分泌以及提高胞浆Ca^2 水平。结论：抗人DAF单抗与抗人CD3单抗可协同刺激人外周血T细胞活化。

Synergistic interaction between decay-accelerating factor and CD3 on activation of human peripheral T lymphocytes

Objective To study the mechanism of T cell activation mediated by DAF which provides a co stimulatory signal. Methods Three McAbs which direct against SCR3, SCR1 and SCR4 of human DAF respectively was used alone or in combination with McAb to human CD3 (UCHT1) to induce the activation of human peripheral T lymphocytes.Results Cross linked anti DAF McAb in combination with McAb to human CD3 can induce the elevation of intracellular calcium, induction of IL 2 secretion, as well as proliferation of T lymphocytes. By contrast, cross linking of CD55 alone by DG3, DG9 and DA11 was failed to cause these events.Conclusion Our study suggests there is a synergistic interaction between DAF and CD3 on activation of human peripheral T lymphocytes.
