远志及其提取物对抑郁小鼠抗抑郁作用研究

摘    要:目的:探讨远志及其提取物对抑郁症模型小鼠的抗抑郁作用。方法:将60只小鼠随机分为空白组、模型组、阳性组、远志原药组、远志多糖组5组,每组12只,雌雄各半。除空白组外,其余各组利用不可预知的长期温和刺激建立慢性应激抑郁模型。空白组及模型组灌胃蒸馏水,阳性组灌胃盐酸氟西汀,远志原药组灌胃远志水煎煮液,远志多糖组灌胃远志多糖提取物,连续24 d。通过强迫游泳实验和悬尾实验评价各给药组的疗效,用试剂盒分别测小鼠脑组织超氧化物歧化酶(SOD)活性和丙二醛(MDA)的含量,以评价药物的作用。结果:强迫游泳和悬尾实验中,与空白组比较,模型组小鼠呈明显抑郁状态,游泳和悬尾不动时间明显延长(P0.05),SOD活性降低(P0.05),MDA含量升高(P0.05)。与模型组比较,各治疗组小鼠游泳和悬尾不动时间均缩短(P0.05),SOD活性升高(P0.05),MDA含量降低(P0.05)。结论:远志及其提取物均有抗抑郁的作用,但远志水煎剂抗抑郁的作用优于远志多糖。

Abstract:Objective:To study the antidepressant effect of polygala and its extracts on depressive mice. Methods:60 mice were divided into blank group,model group,positive group,polygala decoction group and polygala polysaccharide group with 12 mice in each group. In addition to blank group,the others groups were used to establish the chronic stress depres-sive model induced by long-term and unpredictable stimulation. The blank group and model group were given distilled wa-ter,the other groups were given fluoxetine hydrochloridem,polygala decoction and polysaccharide by intragastric adminis-tration respectively for successive 24 days. The effects of each group were evaluated by forced swimming experiment and tail suspension experiment. MDA content and SOD activity were measured to estimate the effect of drugs. Results:Com-pared with blank group,the mice of model group showed obvious state of depression,the immobility time of swimming and tail suspension was significantly extended(P〈0.05),SOD activity was decreased(P〈0.05)and MDA content was increased (P〈0.05)during the experiment of forced swimming and tail suspension. Compared with the model group,the immobility time of swimming and tail suspension of each treatment groups was markedly shortened,SOD activity was increased(P〈0.05), MDA content was decreased(P〈0.05). Conclusion:Polygala and its extracts have antidepressant effect,and the effect of poly-gala decoction prefer to that of polysaccharide.
