亲水性处理喷砂大颗粒酸蚀钛表面对成骨细胞黏附及铺展的影响

摘    要:目的探讨亲水性处理喷砂大颗粒酸蚀(modSLA)钛表面对牙槽骨成骨细胞黏附及铺展行为的影响。方法将人牙槽骨成骨细胞分别接种于喷砂大颗粒酸蚀(SLA)和亲水性modSLA的钛片表面。在接种后1、3、6、12、24 h,利用细胞计数法测定两种钛片表面的细胞黏附率,使用共聚焦激光扫描显微镜观测细胞骨架铺展情况并作荧光积分强度(IOD)定量检测,使用Image-Pro Plus 6.0软件测定各时间点细胞形状因子。结果细胞接种后1、3 h,modSLA组表面细胞黏附率高于SLA组(P<0.05),而6、12、24 h后组间比较差异无统计学意义。接种1 h后,两组细胞形态呈圆形;接种3、6 h后两组细胞逐渐伸展,并形成丝状伪足与材料表面相连。细胞接种12 h后,与SLA相比,modSLA组表面细胞可以观察到较清晰的肌动蛋白结构、细胞铺展更充分。细胞接种3、6、12、24 h后modSLA组IOD值显著高于SLA组(P<0.05)。随时间延长两种材料表面细胞的IOD值均显著增加(P<0.05)。3、6、12 h mod SLA组细胞形状因子均值显著小于SLA组,差异有统计学意义(P<0.05)。随时间延长两种材料表面细胞的形状因子均显著减小,差异有统计学意义(P<0.05)。结论亲水性化学处理能显著促进人牙槽成骨细胞在SLA钛表面的黏附、铺展。

Abstract:Objective To explore the effect of hydrophilic modification of sandblasted,large-grit,acid-etched(modSLA) tianium surface on adhesion and spreading behavior of human alveolar osteoblasts(AOBs).Methods AOBs were inoculated on disks with SLA and modSLA surfaces.Cell attachment rates were assessed by cells counting method at 1,3,6,12 and 24 h after seeding.The cytoskeletal actin organization were observed using confocal laser scanning microsopy(CLSM) and analyzed quantitatively using the integrated optical density(IOD) values at 1,3,6,12 and 24 h.The cell shape factors were evaluated by Image-Pro Plus 6.0 software at each time points.Results modSLA surfaces demonstrated significantly higher attachment rates than SLA ones after 1 and 3 h,while no statistical difference found at 6,12 and 24 h.Cells appeared to spread more,had more numerous and defined actin stress fibres on modSLA at 12 h,and the IOD values on modSLA were significantly higher than those on SLA at 3,6,12,24 h after seeding.The IOD values increased significantly with time(P<0.05) in both material groups.The shape factor in modSLA group was much smaller than the ones in SLA at 3,6,12 h after cell seeding,and the value decreased with time on both materials(P<0.05).Conclusion Hydrophilic modification can promote significantly adhesion and spreading of human AOBs on SLA surface.
