柠檬醛诱导急性早幼粒细胞白血病细胞株NB4凋亡机制的研究

摘    要:目的探讨柠檬醛诱导急性早幼粒细胞白血病细胞株NB4凋亡的机制。方法不同浓度的柠檬醛处理NB4细胞48 h后,RT-PCR法检测凋亡相关基因Bcl-2及Bax的mRNA表达变化,流式细胞仪检测细胞caspase-3及核因子kappaB(NF-κB)表达的变化。结果随着柠檬醛浓度增加,RT-PCR结果显示受测细胞的Bax mRNA表达增强,Bcl-2mRNA表达减弱;流式细胞仪检测结果显示受测细胞的caspase-3表达增加,NF-κB表达下降。结论降低NF-κB、Bcl-2表达及促进Bax、caspase-3表达可能是柠檬醛诱导NB4细胞株凋亡的机制之一。

Abstract:Objective To investigate the mechanism of the apoptosis induced by citral in the acute promyelocytic leukemia cell line NB4.Methods NB4 cells were treated with different concentration of citral for 48 hours,then mRNA expression of apoptosis related gene Bcl-2 and Bax were detected by RT-PCR method,and the expression of caspase-3 and NF-κB were detected by FCM.Results Bax and caspase-3 were up-regulated and Bcl-2 and NFκB were down-regulated in NB4 cells which treated by citral in a dose-dependent manner.Conclusion Inducing the Bax and caspase-3 up-regulation and the Bcl-2 and NF-κB down-regulation may be one of mechanism involved in the apoptosis induced by citral in NB4 cells.
