二苯乙烯苷对沙鼠脑缺血/再灌注损伤的保护作用及部分机制

摘    要:目的研究二苯乙烯苷(TSG)对沙鼠脑缺血/再灌注损伤的保护作用及部分机制。方法采用改良法建立沙鼠脑缺血/再灌注模型。沙鼠随机分为6组,假手术组、模型对照组、TSG大、中、小剂量(6、3、1.5 mg/kg),阳性对照药依达拉奉注射组(3 mg/kg)。7 d后通过Morris水迷宫测定学习记忆功能;脑组织匀浆行超氧化物歧化酶(SOD)、谷胱甘肽过氧化物酶(GSH-Px)活性和丙二醛(MDA)含量测定;免疫组化SABC法显示Nestin蛋白在脑组织中的表达。结果与假手术组相比,模型组游出时间和游泳距离明显延长,而TSG治疗组、依达拉奉对照组较模型组明显缩短(P<0.05,P<0.01)。与假手术组相比,模型组沙鼠脑组织中SOD活性、GSH-Px酶活力单位明显降低,MDA含量明显升高。TSG(6、3 mg/kg)可显著提高血清SOD活性和GSH-Px酶活力单位,TSG(6、3、1.5 mg/kg)可明显降低MDA含量;TSG(6、3、1.5 mg/kg)Nestin蛋白的表达水平较模型组有明显增高。结论 TSG对脑缺血/再灌注损伤有明显的保护作用,其机制可能与抗自由基损伤、抑制体内脂质过氧化水平及促进神经细胞的修复有关。

Effect of tetrahydroxystilbene glucoside on gerbils with cerebral ischemia-reperfusion injury

Abstract:Objective To study the protective and therapeutic effects of tetrahydroxystil-bene glucoside(TSG) on cerebral ischemia-reperfusion injury.Methods The improved method to establish the gerbils cerebral ischemia-reperfusion model.The male gerbils were randomly divided into six groups: sham group,model group,TSG large,medium,small doses(6,3,1.5 mg/kg) groups,the positive control of edaravone medicine injection(3 mg/kg).The function of learning and memory were detected by Morris water maze after 7 days;SOD,GSH-Px activity and MDA contents were detected by brain tissue homogenate;Nestin protein expression was detected by immunohistochemical SABC method.Results Compared with sham group,the time of swimming out and the distance in model group was significantly extended.Compared with model group,the time of swimming out and the distance in TSG treatment group,edaravone control group obviously reduced.Compared with sham group,SOD,GSH-Px enzyme activity reduced significantly in gerbils brains,MDA content was obviously increased in model group.TSG(6,3 mg/kg) could significantly improve the serum SOD activity and GSH-Px enzyme activity,TSG(6,3,1.5 mg/kg) could obviously reduce the MDA content;The Nestin protein expression in TSG(6,3 mg/kg) group was significantly higher than that in model group.Conclusion TSG has positively protective and therapeutic effect on cerebral ischemia-reperfusion injury.Its mechanism may be relevant with free radical damage,inhibit the body of lipid peroxidation and promote the level of nerve cells repair.
