肝郁脾虚因素对小鼠H22腹水瘤细胞周期的影响及疏肝健脾方的作用研究

目的:观察肝郁脾虚因素对H22荷瘤小鼠腹水瘤细胞增殖周期的影响以及疏肝健脾方药的治疗作用。方法:小鼠腹腔接种H22肿瘤细胞后给予肝郁脾虚因素刺激,建立肝郁脾虚证与移植性腹水瘤的病证结合模型,治疗组给予疏肝健脾方药。10d后处死小鼠,抽取腹腔液用流式细胞术检测肿瘤细胞增殖周期。结果:病证结合组与单纯肿瘤组比较,S期细胞比率(S-phasefraction,SPF)明显上升(P<0.01),G1期细胞比率明显下降(P<0.01);中药治疗组与病证结合组比,SPF明显下降(P<0.01),G1和G2/M期细胞比率明显上升(P<0.01)。结论:肝郁脾虚因素可以使H22荷瘤小鼠腹水瘤细胞SPF明显上升,促进肿瘤细胞分裂增殖。而疏肝健脾方药对肝郁脾虚因素所致的肿瘤细胞增殖周期改变有明显的逆转作用。

Fffect of Liver-Stagnation and Spleen-Deficiency on the Cell Cycle of H22 Ascitic Fluid Tumor in Mice and the Treatment of Shuganjianpifang

Objective:To investigate the effect of liver-stagnation and spleen-deficiency on the cell cycle of H22 ascitic fluid tumor in mice and the treatment of Shuganjianpifang. Methods:Tumor-bearing mice were stimulated with the factor of liver-stagnation and spleen-deficiency to establish the model of integrated disease and syndrome (IDS). And the treatment group was given Shuganjianpifang. 10 days later, the ascitic fluid cells of each group were extracted and the cell cycle of them was detected by flow cytometry. Result:SPF increased significantly in IDS group compared with the control (P<0.01). Compared with the IDS group, SPF in the treatment group was diminished, and G2/MSP was enhanced. Difference between the two groups was notable (P<0.01). Conclusion:The factor of liver-stagnation and spleen-deficiency could enhance the SPF of H22 ascitic tumor cells and promote the proliferation of tumor. And Shuganjianpifang had a therapeutic effect on the change of cell cycle in tumor-bearing mice stimulated with liver-stagnation and spleen-deficiency.
