摘    要:为建立一种恙螨和鼠宿主体内恙虫病立党次体检测最敏感特异的基因技术万法,本文对包括DNA抽提、PCR、核酸杂交、PCR结合核酸杂交作了系统比较研究,结果证明,PCR结合核酸杂交比单用PCR敏感10倍,比单独核酸杂交敏感1000倍.而DNA快速抽提可代替酚一氯访法应用于鼠血和病人标本的检测,会抑制DNA快速抽提法能使阳性恙螨PCR检出率提高。

Abstract:In order to establish the most sensitive and specific methods to detect Rickettsia tsutsugamushi in Trombiculid mite and mice. we have made a systematic comparative study on DNA extraction, PCR, hybridization and PCR-hybridization. The resrlt showed the sensitivity of PCR-hybridization was 10 times higher than that of PCR,1000 times higher than that of DNA-DNA hybridization. Quick DNA extaction could be used in detection of mice and patients blood in place of phenol-chloroform. The eliminating inhibition and quick DNA extraction method could improved the detection rate of positive Trombiculid mite.
