摘    要:目的:采用反相高效液相色谱法测定当归挥发油中藁苯内酯含量,为当归挥发油皮肤渗透动力学研究提供依据.方法:以乙腈-水(78∶22)为流动相,流速1.0mL/min,25℃下经Inertsil ODS-3(4.6mm×250mm,5μm)色谱柱分离,于280nm波长处检测.结果:藁苯内酯与挥发油中其它成分的分离效果良好,在0.5~80.0mg/L浓度范围内呈良好的线性关系,回归方程为:Y=45342.52X-8462.62(r=0.9998,n=5);平均回收率为100.07%,相对标准偏差为0.18%.结论:本方法简便、准确,可快速测定当归挥发油中藁苯内酯的含量,可为质量控制提供依据.

Abstract:AIM: To establish a reversed-phase high performance liquid chromatographic ( RP-HPLC ) method for simultaneous determination of ligustilide in the essential oil of Angelica sinensis (Oliv.) Diels. METHODS: The mobile phase was acetonitrilewater(78:22) and the flow rate was about 1 mL/min. Inertsil ODS-3(4.6 mm × 250 mm, 5 μm) column was used and the detective wavelength was 280 nm. RESULTS: The calibration curves were linear in the concentration range of 0. 5 - 80. 0 mg/L for ligustilide. The regression equation was Y = 45 342.52 X - 8462.62 ( r = 0. 9998, n = 5 ) , the average recovery was 100.07% and the relative standard deviations (RSD) was 0.18%. CONCLUSION: Our established method is simple,quick and accurate for the simultaneous separation and determination of ligustilide in the essential oil of Angelica sinensis ( Oliv. ) Diels.
