| 摘 要： | 目的:探讨宫颈癌细胞株中survivin mRNA表达分子信标荧光显像的检测。方法:体外培养宫颈癌细胞株HeLa和SiHa,滴加100nM的survivin mRNA分子信标,荧光显微镜下观察荧光强度,并用western blot及免疫组化方法对其表达进行验证。结果:两种Survivin mRNAMB在HeLa和SiHa中观察到了强荧光信号,且SiHa荧光信号强于Hela。结论:分子信标技术可成功检测宫颈癌细胞survivin mRNA的表达,为宫颈癌的早期诊断提供了又一可能的手段。

| Abstract: | Objective:To study molecular beacon imaging detection which was expressed by survivin mRNA in cervical cancer cell lines.Methods:Human cervical cancer cell lines(HeLa and SiHa)were cultured in vitro,and the fluorescent signals were detected under fluorescent microscope,after adding 100nM surviving mRNA molecular beacon.The results were verified by western blot and immunohistochemistry.Results:Two kinds of survivin mRNA molecular beacon had strong fluorescent signals in Hela and SiHa cervical cancer cell lines,which was stronger in SiHa cell line.Conclusion:The technique of molecular beacon imaging can successfully detect the expression of survivin mRNA in cervical cancer cells,which provides an alterative way for early diagnosing of cervical cancer. |
