解虑安神汤抗焦虑作用与GABAA-R关系的初步研究

目的:观察GABAA-R拮抗剂荷苞牡丹碱(Bicuculline,Bic)及苯二氮(BDZ)受体拮抗剂氟马西尼(Flumazenil,Flu)对解虑安神汤(JLAST)乙酸乙酯提取部位(J-A)和正丁醇提取部位(J-B)抗小鼠焦虑行为的影响,研究JLAST抗焦虑作用与GABA-BDZ-Cl-复合体的关系.方法:小鼠连续给予J-A、J-B 7 d,于末次给药后1 h,脑室内分别注射Bic或Flu,观察J-A、J-B对小鼠高架十字迷宫实验行为学的影响.结果:与空白对照组相比,①地西泮组、J-A组、J-B组均可增加小鼠进入开臂次数、进入开臂次数百分比(OE%)及开臂停留时间百分比(OT%),差异有显著性意义;②地西泮+Bic组、J-A+Bic组、J-B+Bic组对小鼠进入开臂次数、在开臂停留时间及OE%、OT%无明显影响,差异无显著性意义;③地西泮+Flu组、J-A+Flu组对小鼠进入开臂次数、在开臂停留时间及OE%、OT%无明显影响,差异无显著性意义;J-B+Flu组可增加小鼠进入开臂次数、在开臂停留时间百分比(OT%),缩短在闭臂停留时间,差异有显著性意义.结论:JLAST抗焦虑的机制与GABAA-R有关,其中J-A可能通过GABA-BDZ-Cl-复合体途径发挥作用,J-B则可能通过GABAA-R相关的其他途径发挥作用.

A Preliminary Study on the Relation between the Anxiolytic Effects of Jielv Anshen Decoction and GABAA
