骆驼蓬总碱致突变性实验研究

摘    要:目的研究骆驼蓬总碱的致突变性,为其临床安全用药提供依据。方法进行小鼠伤寒沙门菌微粒体酶(Ames)试验,设4个剂量,分别为40、200、1000、5000彬皿;小鼠骨髓嗜多染红细胞微核试验,设3个剂量组,分剐为229.68、114.84、57.42mg/kg,另各设1个阴性和阳性对照组;小鼠精子畸形试验,剂量设置同微核试验,观察骆驼蓬总碱的致突变作用;结果3项致突变试验结果均为阴性,表现为:(1)Ames试验各组在加与不加S-9的情况下,其回变菌落数均未超过其自发回变菌落数的2倍;(2)骆驼蓬总碱3个剂量组对小鼠骨髓微核率的影响。与对照组比较差异无显著性意义(P〉0.05);(3)骆驼蓬总碱各刺量组对精子畸形率的影响与对照组比较差异亦无显著性意义(P〉0.05);结论在本实验条件下,骆驼蓬总碱对受试动物未见有致突变性毒性反应发生。

Abstract:Objective To evaluate mutagenicity of Harmala alkaloids, and provide basis for safe clinical application. Methods Ames test was used in this study with four dosage of 40,200,1000,5000μg/lm, Micronueleus test of bone marros in mice was used in this study with three dosage of 229.68,114.84,57.42 mg/kg. There were another negative controlling group and positive controlling group. Mutagenicity of Harmala alkaliods was observed through mice sperm deformation test. Results Three mutagenicity tests all had negative result. ( 1 ) Results of Ames test showed that number of colony with back mutation with or without S - 9 did not exceed the double of the number of those with spontaneous mutation. (2) There was no significant difference on mice micronuleus rate of bone marrow between three dosage groups of Harmala alkaloids and controlling group( P 〉 0.05 ), (3) There is no significant difference between Harmala alkaloid and controlling group on sperm deformation rate. Conclusion Under condition of this experiment, there was no mutagenicity to animal subjects from harmala alkaloids.
