辛伐他汀对动脉粥样硬化大鼠主动脉血红素氧合酶-1表达的影响

摘    要:目的探讨辛伐他汀对动脉粥样硬化(AS)大鼠主动脉血红素氧合酶1(HO-1)表达的影响。方法取健康SD大鼠24只,随机分为3组;对照组、AS模型组和治疗组,每组8只。AS模型组和治疗组在实验开始时,一次性腹腔注射维生素D3 60万U/kg,以后喂食高脂饮食;对照组喂食普通饲料。4周后治疗组灌胃给予辛伐他汀10mg/(kg·d)治疗。干预治疗4周后处死大鼠,取主动脉观察形态及病理学变化,同时采用免疫组织化学方法检测大鼠主动脉HO-1的表达情况,并应用人工计数分析法对免疫组化结果进行定量分析。结果(1)AS模型组主动脉出现典型AS斑块,其主动脉HO-1的表达水平显著高于对照组(P〈0.001)。(2)治疗组的主动脉粥样硬化程度与AS模型组比较明显减轻,但其HO-1的表达水平显著高于AS模型组(P〈0.001)。结论(1)辛伐他汀能够上调HO-1在AS大鼠主动脉的表达水平。(2)辛伐他汀可能通过上调HO-1的表达而发挥抗AS作用。

Abstract:Objective To investigate the effect of simvastation on the expression of heme oxygenase-1(HO-1) in the aortic artery of atherosclerotic(AS) rat models.Methods Twenty-four SD rats were assigned into 3 groups randomly:normal control group,AS model group and simvastatin treatment group (8 rats in each group).The rats in AS model group and treatment group were injected with a single dose of vitamin D3(600 000 U/kg)and fed with high fat diet.Basic food was given in normal control group.The rats in treatment group were treated with simvastatin by gastric tube 4 weeks later.All of the rats were sacrificed at the end of 4 weeks' treatment and the aortic artery was observed by light microscopy.The expression of HO-1 protein in the aortic artery of AS rat models was investigated by immunohistochemistry and manual work analysis.Results (1)Typical AS plaques were seen in the aorta of the model group.The expression level of HO-l protein were significantly higher in the AS model group than that of control group(P<0.001).(2)The atheroselotic change of aorta in treatment group was much mild than that of the AS mode1 group.The expression level of HO-1 protein was signiricantly higher in the treatment group than that of AS model group(P<0.001).Conclusion (1)Simvastatin may be able to up-regulate the expression level of HO-l protein in the aortic artery of AS rat models.(2)The anti-atherosclerosis of simvastatin maybe occur through up-regulation of the expression of HO-l protein.
