5-HT3受体在慢性神经病理性疼痛模型脊髓组织中的表达

摘    要:目的观察慢性神经病理性疼痛大鼠脊髓组织中5-HT,受体的表达变化。方法采用大鼠坐骨神经结扎(慢性压迫性损伤)制备慢性神经病理性疼痛模型。坐骨神经结扎造模后7天,采用测定热缩足反射潜伏期(TWL)和机械缩足反射阀值(MWT)2种行为学方法检测大鼠痛觉闽值变化,免疫组织化学染色法检测大鼠脊髓背角5-HT,受体表达,Westernblot方法检测大鼠脊髓背角中5-HT,受体蛋白水平。结果造模7天后:模型组大鼠的TWL和MWT均低于正常组和假手术组(P〈0.05),模型组大鼠脊髓背角组织中5-HT,受体表达明显高于正常组和假手术组(P〈0.05)、5-HT,受体蛋白水平高于正常组和假手术组(P〈0.05)。结论在慢性神经病理性疼痛中,脊髓背角组织中5-HT,受体表达增加;脊髓背角组织中5-HT,受体可能参与了慢性神经病理性疼痛的痛觉信息传导过程。

Abstract:Objective To investigate the expression of 5 - HT3 receptor in the spinal cord tissue of rats with chronic neuropathic pain. Methods Chronic neuropathie pain model was established by sciatic nerve ligation ( chronic constric-tion injury). After model replication in the 7th day, thermal withdrawal latency (TWL) and mechanical withdrwal threshold (MWT) were used to detect the pain threshold of the rats. Immunohistochemieal staining was used to detect the expression 5 - HT3 receptors in the spinal dorsal horn. Western blot was used to detect the protein level 5 - HT3 receptors in the spinal dorsal horn. Results After model replication in the 7th day: ①The pain threshold by TWL and MWT of model rats were lower than that of both control and sham operation group ( P 〈 0.05 ) ; ②The expression of 5 - HT3 re-ceptor in the spinal dorsal horn of model rats was higher than that of both control and sham operation group ( P 〈 0.05 ) ;③The protein level of 5 - HT3 receptor in the spinal dorsal horn of model rats was higher than that of both control and sham operation group ( P 〈 0.05 ). Conclusion The expression of 5 - HT3 receptors is increased in the spinal dorsal horn on chronic neuropathic pain, and 5 - HT3 receptor in the spinal dorsal horn may involve in the conductive process of pain.
